Cell-induced confinement effects in soft tissue mechanics

چکیده

The mechanical properties of tissues play a critical role in their normal and pathophysiological functions such as tissue development, aging, injury, disease. Understanding mechanics is important not only for designing realistic biomimetic materials engineering drug testing but also developing novel diagnostic techniques medical interventions. Tissues are heterogeneous consisting cells confined within extracellular matrices (ECMs), both which derive structural integrity, at least part, from networks biopolymers. However, the rheology purified reconstituted biopolymer fails to explain many key aspects mechanics. Notably, typically soften under applied compression, whereas soft like liver, fat, brain instead stiffen when compressed. While continuum models can readily capture this compression-stiffening behavior, underlying mechanism fully understood. In perspective paper, we discuss several recently proposed microscopic mechanisms that may compression stiffening tissues. These include (I) interactions between ECM volume-preserving inclusions promote extension-dominated fibrous ECMs subject uniform (II) with rigid non-uniform (III) other internal physical constraints cause ECMs, (IV) propagation compressive forces through jammed, cells. We further identify few open problems understanding structure–function relationship soft-tissue

Mechanics of Flexible Needles Robotically Steered through Soft Tissue

The tip asymmetry of a bevel-tip needle results in the needle naturally bending when it is inserted into soft tissue. This enables robotic needle steering, which can be used in medical procedures to reach subsurface targets inaccessible by straight-line trajectories. However, accurate path planning and control of needle steering requires models of needle-tissue interaction. Previous kinematic m...

Confinement Effects on Phase Behavior of Soft Matter Systems

When systems that can undergo phase separation between two coexisting phases in the bulk are confined in thin film geometry between parallel walls, the phase behavior can be profoundly modified. These phenomena shall be described and exemplified by computer simulations of the Asakura-Oosawa model for colloidpolymer mixtures, but applications to other soft matter systems (e.g. confined polymer b...

Soft Confinement for Polymer Solutions

As a model of soft confinement for polymers, we investigated equilibrium shapes of a flexible vesicle that contains a phase-separating polymer solution. To simulate such a system, we combined the phase field theory (PFT) for the vesicle and the self-consistent field theory (SCFT) for the polymer solution. We observed a transition from a symmetric prolate shape of the vesicle to an asymmetric pe...

Inverse tissue mechanics of cell monolayer expansion

Living tissues undergo deformation during morphogenesis. In this process, cells generate mechanical forces that drive the coordinated cell motion and shape changes. Recent advances in experimental and theoretical techniques have enabled in situ measurement of the mechanical forces, but the characterization of mechanical properties that determine how these forces quantitatively affect tissue def...
