Cell-induced confinement effects in soft tissue mechanics

The mechanical properties of tissues play a critical role in their normal and pathophysiological functions such as tissue development, aging, injury, disease. Understanding mechanics is important not only for designing realistic biomimetic materials engineering drug testing but also developing novel diagnostic techniques medical interventions. Tissues are heterogeneous consisting cells confined within extracellular matrices (ECMs), both which derive structural integrity, at least part, from networks biopolymers. However, the rheology purified reconstituted biopolymer fails to explain many key aspects mechanics. Notably, typically soften under applied compression, whereas soft like liver, fat, brain instead stiffen when compressed. While continuum models can readily capture this compression-stiffening behavior, underlying mechanism fully understood. In perspective paper, we discuss several recently proposed microscopic mechanisms that may compression stiffening tissues. These include (I) interactions between ECM volume-preserving inclusions promote extension-dominated fibrous ECMs subject uniform (II) with rigid non-uniform (III) other internal physical constraints cause ECMs, (IV) propagation compressive forces through jammed, cells.
